   Function Description

By screening the downstream genes of MALAT1, we noticed that microRNA 203(miR203), an oncogene suppressor in numerous cancers, is inversely correlated to both MALAT1 and DCP1A expressions. To measure the effects on drug resistance, after 48 hours of transfection with either siRNAs or plasmids, the cells were seeded and cultured with a density of 5,000 cells/well in a flatbottomed 96-well plate in 10%FBS mixed with cisplatin(Sigma) at a concentration of 20ug/ml for consecutive 48 h. 20uL of 3-(4, 5-Dimethyl-2-thiazolyl)-2, 5-diphenyl-2Htetrazolium bromide (MTT) (Sigma) (5 mg/ml) was applied at 12, 24, 36, 48 hours and incubated for 4 h. Kaplanā??Meier survival curve analysis showed that the overall survival time of CRC patients with higher DCP1A expression was significantly shorter than patients with lower DCP1A levels (log-rank test, *P < 0.05).These results proposed a new molecular mechanism of MALAT-miR203-DCP1A axis which is involved with the development and contributes to the malignancy of colorectal cancers.
